http://www.ncbi.nlm.nih.gov/pmc/articles/PMC3624207/
Tulee mieleen  "akuutti monosyyttileukemia"- Ebolan suhde monosyyteihin, niiten rekrytoimiseen on ylläolevassa artikkelissa. Muta tässä aola ensin otan vertauskohdaksi histiosytoosin ja siten katson missä ebola eroaa  alalolevasta histiosytoosityypistä.

Myös  Lintuvirus H5N1 pahimmillaan voi aiheuttaa erään sortin HLH , hemofagosyyttistä lymfohistiosytoosin kaltaista tilaa  Samoin muut virukset.

Minkälainen se on? http://www.bloodjournal.org/content/118/15/4041?sso-checked=true

Wikipedia kertoo  lyhyesti: mm:

Malignant histiocytosisa Malignant histiocytosis (also known as "Histiocytic medullary reticulosis"[1]:744) is a rare hereditary disease ....characterized by histiocytic infiltration of the lungs and lymph nodes. The liver, spleen, and central nervous system can also be affected. Histiocytes are a component of the immune system that proliferate abnormally in this disease. In addition to its importance in veterinary medicine, the condition is also important in human pathology.[2]
Histiocytic disorders...
A histiocyte is a differentiated tissue cell that has its origin in the bone marrow. The source for histiocytes is the monocyte/macrophage line. Monocytes (found in the blood) and macrophages (found in tissue) are responsible for phagocytosis (ingestion) of foreign material in the body. Langerhans cells are dendritic cells found in the skin and function by internalizing antigens (foreign particles) and presenting them to T cells. They arise from monocytes.[3] Histiocytic disorders refer to diseases that are caused by abnormal behavior of these cells. They include the following:...

Farletuzumab

LÄHDE: http://mericaltinoz.blogspot.se/2014/10/ebola-folate-receptor-1-farletuzumab.html

Sitaatti:
EBOLA, a virus disease causing a global health alarm and one of the most fatal type of viral hemorrhagic fevers employs folate receptor-1 for cellular entry. Folate receptor-1/FLR-1 is also one of the important proteins upregulated in cancer facilitating enhanced folate entry, which is used for nucleotide synthesis to maintain increased DNA synthesis. Currently, FLR-1 inhibitor farletuzumab is tested in Phase-3 trials for the treatment of ovarian cancer. This agent may also act beneficial in blocking EBOLA entry into cells and help alleviating the severity of EBOLA. Vice versa, if medicine could modify the deadly course of this virus disease by modifying the viral genotype in future, EBOLA virus may act as a gene therapy vector similar to the current approaches to employ modified AIDS virus and other members of lentiviridae in gene delivery.

Kuitenkin on olemassa lääke FLR-1 inhibiittori, farletuzumab jota onkologiassa  Ca ovarii hoidossa tutkitaan III faasissa.

Human infection with avian influenza A(H7N9) virus – China

Disease outbreak news
18 November 2014
On 15 November 2014, the National Health and Family Planning Commission (NHFPC) of China notified WHO of 3 additional laboratory-confirmed cases of human infection with avian influenza A(H7N9) virus, including 1 death.

Details of the cases are as follows

  • A 54-year-old female from Shihezi City, Xinjiang Uyghur Autonomous Region who developed symptoms on 19 October. The patient died on 1 November. Exposure to live poultry is unknown.
  • A 58-year-old female from Changzhou City, Jiangsu Province who developed symptoms on 24 October. The patient was admitted to a hospital on 28 October. She is now in critical condition. The patient has a history of exposure to a live poultry market.
  • A 45-year-old female from Changzhou City, Jiangsu Province who developed symptoms on 3 November. The patient was hospitalized on 5 November. Her current condition is mild. The patient has a history of exposure to a live poultry market.

The Chinese Government has taken the following surveillance and control measures

  • Strengthen surveillance and situation analysis
  • Reinforce case management and medical treatment
  • Conduct risk communication with the public and release information.
WHO continues to closely monitor the H7N9 situation and conduct risk assessment. So far, the overall risk associated with the H7N9 virus has not changed.

WHO advice

WHO advises that travellers to countries with known outbreaks of avian influenza should avoid poultry farms, or contact with animals in live bird markets, or entering areas where poultry may be slaughtered, or contact with any surfaces that appear to be contaminated with faeces from poultry or other animals. Travellers should also wash their hands often with soap and water. Travellers should follow good food safety and good food hygiene practices.
WHO does not advise special screening at points of entry with regard to this event, nor does it currently recommend any travel or trade restrictions. As always, a diagnosis of infection with an avian influenza virus should be considered in individuals who develop severe acute respiratory symptoms while travelling or soon after returning from an area where avian influenza is a concern.
WHO encourages countries to continue strengthening influenza surveillance, including surveillance for severe acute respiratory infections (SARI) and to carefully review any unusual patterns, in order to ensure reporting of human infections under the IHR (2005), and continue national health preparedness actions.

Comprehensive functional analysis of N-linked glycans on Ebola virus GP1.
Suomennosta tiivistelmästä, Abstract

Ebolaviruksen sisäänpääsy soluun vaatii virion pinnan glykoproteiinin apua. GP on koostunut treimeeristä joaas on heterodimeerejä GP1/GP2. (peplomeeri).
GP1 alayksikössä on kaksi hyvin vahvasti glykosyloitnutta domeenia, glykaani Gap ja musiinin kaltainen domaani (MLD)
Ebola virus (EBOV) entry requires the virion surface-associated glycoprotein (GP) that is composed of a trimer of heterodimers (GP1/GP2). The GP1 subunit contains two heavily glycosylated domains, the glycan cap and the mucin-like domain (MLD).  The glycan cap contains only N-linked glycans, whereas the MLD contains both N- and O-linked glycans.

Site-directed mutagenesis was performed on EBOV GP1 to systematically disrupt N-linked glycan sites to gain an understanding of their role in GP structure and function.
  • Kaikki 15 N-glykosylaatiokohtaa  voitiin poistaa  aiheuttamatta GP1 molekyylin kompromittoitumsita.
All 15 N-glycosylation sites of EBOV GP1 could be removed without compromising the expression of GP. The loss of these 15 glycosylation sites significantly enhanced pseudovirion transduction in Vero cells, which correlated with an increase in protease sensitivity. Interestingly, exposing the receptor-binding domain (RBD) by removing the glycan shield did not allow interaction with the endosomal receptor, NPC1, indicating that the glycan cap/MLD domains mask RBD residues required for binding.
  • Sitten testattiin vaikutusta kalsiumista riippuviin C-lektiineihin CLEC.
The effects of the loss of GP1 N-linked glycans on Ca(2+)-dependent (C-type) lectin (CLEC)-dependent transduction were complex, and the effect was unique for each of the CLECs tested.
  •  EBOV viruksen pääsy hiiren vatsaontelon makrofageihin riippui GP1 N-glykaanista, mikä viittaisi siihen että CLEC-GP1 N-glykaanin vuorovaikutusta ei tarvitaisi tähän tärkeään primääriin soluun pääsyyn.
Surprisingly, EBOV entry into murine peritoneal macrophages was independent of GP1 N-glycans, suggesting that CLEC-GP1 N-glycan interactions are not required for entry into this important primary cell.
  •  Lopuksi tutkijat  selvittivät miten  GP1 N-glykaanien  poisto musiininkaltaisen alueen  ulkopuolelta  tehosti  antiseerumi- ja  vasta-aineherkkyyttä.  Yhteenvetona tulokset osoittivat , että konservoitunut N-linkkiytynyt glykaanikilpi EBOV-GP1 ydinrakenteessa suojelee vasta-aineitten neutraloivalta vaikutukseslta vaikkakin siitä seuraa virukselle negatiivisesta  vaikutuksesta   soluun sisäänpääsyn tehokkuuden suhteen.
Finally, the removal of all GP1 N-glycans outside the MLD enhanced antiserum and antibody sensitivity. In total, our results provide evidence that the conserved N-linked glycans on the EBOV GP1 core protect GP from antibody neutralization despite the negative impact the glycans have on viral entry efficiency.

Mikä on asian merkitys? IMPORTANCE:

Filovirus outbreaks occur sporadically throughout central Africa, causing high fatality rates among the general public and health care workers. These unpredictable hemorrhagic fever outbreaks are caused by multiple species of Ebola viruses, as well as Marburg virus. While filovirus vaccines and therapeutics are being developed, there are no licensed products. The sole viral envelope glycoprotein, which is a principal immunogenic target, contains a heavy shield of glycans surrounding the conserved receptor-binding domain. We find that disruption of this shield through targeted mutagenesis leads to an increase in cell entry, protease sensitivity, and antiserum/antibody sensitivity but is not sufficient to allow virion binding to the intracellular receptor NPC1. Therefore, our studies provide evidence that filoviruses maintain glycoprotein glycosylation to protect against proteases and antibody neutralization at the expense of efficient entry. Our results unveil interesting insights into the unique entry process of filoviruses and potential immune evasion tactics of the virus.

J Virol. 2012 Jul;86(14):7473-83. doi: 10.1128/JVI.00136-12. Epub 2012 May 9. Ebolavirus requires acid sphingomyelinase activity and plasma membrane sphingomyelin for infection.
Abstract (Tiivistelmä)

Acid sphingomyelinase (ASMase) converts the lipid sphingomyelin (SM) to phosphocholine and ceramide and has optimum activity at acidic pH. Normally, ASMase is located in lysosomes and endosomes, but membrane damage or the interaction with some bacterial and viral pathogens can trigger its recruitment to the plasma membrane.
  •  Infektoimisensa  varhaisvaiheisiin  rinovirukset ja tuhkarokkovirukset  vaativat  happaman sfingomyelinaasin aktiivisuutta
Rhinovirus and measles viruses each require ASMase activity during early stages of infection.
Both sphingomyelin and ceramide are important components of lipid rafts and are potent signaling molecules. Each plays roles in mediating macropinocytosis, which has been shown to be important for ebolavirus (EBOV) infection.
Here, we investigated the role of ASMase and its substrate, SM, in EBOV infection. The work was performed at biosafety level 4 with wild-type virus with specificity and mechanistic analysis performed using virus pseudotypes and virus-like particles.
We found that virus particles strongly associate with the SM-rich regions of the cell membrane and depletion of SM reduces EBOV infection.
  •  Happamalle sfingomyelinaasientsyymille spesifiset lääkkeet ja multippelit siRNA.t  estivät vahvasti EBOV infektiota ja EBOV glykoproteiineilla  pseudotyypattuja viruksia, mutta eivat estäneet  prototyyppiviruksen  VSV glykoproteiinilla pseudotyypattuja viruksia.
ASM-specific drugs and multiple small interfering RNAs strongly inhibit the infection by EBOV and EBOV glycoprotein pseudotyped viruses but not by the pseudotypes bearing the glycoprotein of vesicular stomatitis virus.
  • Mielenkiintoinen löytö oli virusten kaltaisten partikkelien (VLP)  vahva   sitoutuminen pinnalle lokalisoituneeseen  happamaan sfingomyelinaasiin ja samoin  sfingomyeliinipitoisiin  kohtiin.  Tutkijoitten  mukaan solujen tehokkaalle EBOV - infektoitumiselle  on välttämätöntä happaman sfingomyelinaasin aktiivisuus ja sfingomyeliinin läsnäolo. Tämän tien esto saattaneisi tarjota uusia  väyliä lääketerapiaan.
Interestingly, the binding of virus-like particles to cells is strongly associated with surface-localized ASMase as well as SM-enriched sites. Our work suggests that ASMase activity and SM presence are necessary for efficient infection of cells by EBOV.
The inhibition of this pathway may provide new avenues for drug treatment.

hakusnana: EBOV ja lysosomi? Lektiinit. DC-SIGN/R ja LSECtiini

Interactions of LSECtin and DC-SIGN/DC-SIGNR with viral ligands: Differential pH dependence, internalization and virion binding.

The calcium-dependent lectins DC-SIGN and DC-SIGNR (collectively termed DC-SIGN/R) bind to high-mannose carbohydrates on a variety of viruses. In contrast, the related lectin LSECtin does not recognize mannose-rich glycans and interacts with a more restricted spectrum of viruses. Here, we analyzed whether these lectins differ in their mode of ligand engagement.

LSECtin and DC-SIGNR, which we found to be co-expressed by liver, lymph node and bone marrow sinusoidal endothelial cells, bound to soluble Ebola virus glycoprotein (EBOV-GP) with comparable affinities. Similarly, LSECtin, DC-SIGN and the Langerhans cell-specific lectin Langerin readily bound to soluble human immunodeficiency virus type-1 (HIV-1) GP.
However, only DC-SIGN captured HIV-1 particles, indicating that binding to soluble GP is not necessarily predictive of binding to virion-associated GP. Capture of EBOV-GP by LSECtin triggered ligand internalization, suggesting that LSECtin like DC-SIGN might function as an antigen uptake receptor.
However, the intracellular fate of lectin-ligand complexes might differ. Thus, exposure to low-pH medium, which mimics the acidic luminal environment in endosomes/lysosomes, released ligand bound to DC-SIGN/R but had no effect on LSECtin interactions with ligand. Our results reveal important differences between pathogen capture by DC-SIGN/R and LSECtin and hint towards different biological functions of these lectins.
